細菌由来ADPリボシル化毒素の作用機構と基質特異性

多くの細菌は外毒素を用いて我々宿主の細胞のアクチン細胞骨格系に影響を与え、自分の生存に活かしていると考えられる。特にこの仲間にADPリボシル化毒素は多い。ADPリボシル化毒素を基質の違いで大きく分けると4つのタイプに分類でき、この内Type IIIとType IVの毒素はアクチン細胞骨格系に影響を及ぼす。(この分類は少し古く、今ではもっと多くの基質に対する毒素が見つかっているが、典型的なADPリボシル化毒素を考える上で役に立つ)Type IIIはRhoAを修飾するC3毒素、Type IVはアクチンを修飾するIota毒素(Ia)である。C3毒素はAsn、 IaはArgと修飾されるアミノ酸も異なる。この違いに興味を持ち、構造と機能の研究を始めた。

2003年我々はウェルシュ菌のIaのX線結晶構造解析を行った(JMB 2003 Tsuge et al.)。結晶構造解析により補酵素NADとの結合が見えてくる。これにより、反応機構が議論できる。多くの海外の研究室でも毒素単体の構造は解析されてきた。しかし何かが足りない。反応を考える上で必要なのは基質となるタンパク質である。例えばプロテアーゼの構造研究の場合、一般に基質がわかれば、その基質類似のペプチドとの複合体構造が期待できる。一方、ADPリボシル化毒素の場合は、基質となるペプチドはなく、その基質は宿主のタンパク質全体である。このADPリボシル化毒素と基質タンパク質複合体構造を明らかにしたいと思い、さらに研究を続けた。

我々のグループが明らかした複合体はType IVのIaとアクチン複合体(PNAS 2008 Tsuge et al., PNAS 2013 Tsurumura et al.)、およびType IIIのC3とRhoAの複合体(JBC 2015 Toda et al.)である。特にType IVとType IIIの毒素の構造は非常に似ているが、その基質は全く異なり、修飾するアミノ酸も異なっている。明らかにした基質特異性、いかにタンパク質を認識するか、そして修飾アミノ酸を決定しているかについて紹介したい。

「インフルエンザウイルスとその活性化酵素」

インフルエンザウイルスは、粒子表面に二種類のスパイクタンパク質(ヘマグルチニン HAとノイラミニダーゼ NA)を持っている。HAは、受容体への結合とウイルスエンベロープと細胞膜との融合を担う感染に必須のタンパク質である。ただし、HAが膜融合能を発揮するためには、宿主のプロテアーゼによって、特定のアミノ酸配列部位で開裂される必要がある。すなわち、宿主のプロテアーゼが、インフルエンザウイルスの増殖や病原性に深く関与している。高病原性鳥インフルエンザウイルスとは、そのアミノ酸配列部位の変異によって、プロテアーゼ特異性が変化したウイルスである。最近、私たちは、呼吸器上皮に発現しているプロテアーゼTMPRSS2こそが、季節性インフルエンザウイルスや近年中国で問題になっているH7N9亜型鳥インフルエンザウイルスのHAを開裂する宿主プロテアーゼであることを明らかにした。TMPRSS2遺伝子をノックアウトしたマウスの生体内では、インフルエンザウイルスは感染性を獲得しないため、致死量の数百倍のインフルエンザウイルスを接種しても、そのマウスは症状すら示さない。本セミナーでは、インフルエンザや、その他の呼吸器ウイルス感染症におけるTMPRSS2の意義について紹介する。

[要旨]  The discovery of cytochrome P450 (P450) enzymes began with individuals studying the metabolism of drugs, steroids, and carcinogens, as well as basic scientists interested in unusual hemoproteins. Today the 57 human P450 genes can be categorized on the basis of their substrates, including sterols, xenobiotics, eicosanoids, fatty acids, and fat soluble vitamins (A, D, E, K). Some P450s are still not characterized with regard to their substrates and are considered “orphans.” Deficiencies in the P450s involved in steroid and vitamin metabolism can cause serious health problems. The understanding of the xenobiotic-metabolizing P450s has greatly facilitated drug development and the prediction of bioavailability and potential drug-drug interactions.

Research on vector and vector borne infectious diseases

Vector-borne infectious diseases, is one of the emerging and re-emerging infectious diseases in Sri Lanka as similar as other tropical countries. Although Sri Lanka has reached near elimination levels in Malaria and Lymphatic Filariasis, Dengue and Leishmaniasis, Rickettsiosis continues to be a challenge. Most of the vector borne infections in human are zoonoses, thus as veterinarians are the key persons to control these infections. Veterinary science is a multidisciplinary subject, which includes research on diagnosis, control, prevention and treatment of animal diseases. All animal sciences essentially affect human health either directly or indirectly. Veterinary science protects the human health and well-being by preventing and controlling emerging infectious zoonoses. More than half of the human diseases are animal originated, cause multi host pathogens. Rickettsiosis, leptospirosis, dengue infection and leishmaniasis are major emerging and re-emerging vector borne and zoonotic infectious diseases in Sri Lanka.

Rickettsial organisms infect humans causing a wider array of clinical features and have re-emerged in Sri Lanka where three known disease entities; spotted fever group, murine typhus and scrub typhus do exist. These diseases cause clinical illnesses varying from mild febrile illness to severe multiple organ involvement even leading to fatal outcomes when there is a delay in diagnosis. Hard ticks, Amblyomma sp are possible vector in Sri Lanka. However, reservoir animals for the ricketssial infection in human are still to be identified. Leishmaniasis is an emerging public health problem since late 2000 in Sri Lanka and present a new case appeared every day. At the beginning, the disease was predominantly among the armed forces personnel. Today almost all cases are from civilians indicating further spreading the disease within the district. Since the causative organism of leishmaniasis in Sri Lanka is identified as L. donovani, causative organism of visceral leishmaniasis (VL: Kala Azar) in India, though different zymodeme, the possibilities of visceralising the parasite causing VL cannot be excluded. As veterinarians, we have more responsibility to identify the reservoir animals and mode of transmission through specific vector species. Therefore, we are conducting research on the parasite, the vector, and reservoir host/s, so that a viable control strategy can be devised on firm scientific foundation. Dengue fever is an arboviral infection transmitted by mosquitoes of the genus Aedes. In Sri Lanka, dengue fever epidemics have been occurring with increased magnitudes but the worst epidemic was reported in 2009 with 35008 cases and 346 deaths of which 6638 cases and 51 deaths reported in the Central Province of Sri Lanka. Thus under one health concepts as a veterinarians, and academics, we are responsible on vector borne and zoonotic diseases as WHO report 83% of infectious disease are zoonosis and new zoonosis infection re-emerging and emerging in the worlds.

Siderophore-mediated iron acquisition in Campylobacter

Iron acquisition is critical for bacterial pathophysiology and thus has been proposed as attractive targets for iron-dependent pathogen control. The high-affinity iron acquisition mediated by siderophores is the most efficient and common iron scavenging mechanism in Gram-negative bacteria. Campylobacter has emerged as the leading bacterial cause of foodborne human diseases in many industrialized countries. While Campylobacter does not produce any siderophores, siderophore piracy is evident in various Campylobacter species. To date, enterobactin (Ent), a triscatecholate siderophore with the highest affinity for ferric iron, is the only known physiologically relevant siderophore utilized by Campylobacter for in vivo colonization. The high affinity Ent-mediated iron scavenging is tightly linked to Campylobacter pathogenesis; inactivation of ferric Ent (FeEnt) receptor CfrA or CfrB dramatically reduced and even abolished intestinal colonization by Campylobacter (> 5 log units reduction). Our recent studies have revealed novel features of FeEnt acquisition systems in Campylobacter, and provided new insights into the evolution of FeEnt acquisition in different Campylobacter species. These studies also suggested that Campylobacter is an ideal model organism to examine key issues of Ent-mediated iron scavenging due to its small genome (1.6-1.8 Mbp), inability to synthesize any siderophores, low redundancy of iron acquisition systems, and enormous strain diversity. Salmochelin, the glucosylated enterobactin, is likely another significant iron source for Campylobacter during infection. Various vaccination strategies have been developed and evaluated by targeting FeEnt receptor CfrA; however, these approaches did not provide significant protection against Campylobacter colonization in the intestine. Recently, we have started to explore innovative technology of iron-dependent control. Specifically, using Campylobacter as a model organism, we will use Ent specific antibodies as lipocalin-like agent to inhibit catecholates-mediated iron uptake and starve pathogen out of iron. Evaluation of the in vitro and in vivo efficacy of Ent antibodies in our ongoing studies is expected to result in major conceptual advances in the development of new vaccine and therapeutics to control effectively control the infections caused by Campylobacter and likely other Gram-negative pathogens.
